Peran Asam Askorbat Terhadap Proliferasi dan Diferensiasi Sel Punca Mesenkim Menjadi Osteoblas dan Kondrosit Secara In Vitro

Stem cells are self-renewal cells. They have no specific form and function (undifferentiated) but have the ability to differentiate into other cells. The main factor determining the success of proliferation and differentiation of mesenchymal stem cells in vitro is the culture condition. Mesenchymal stem cells have a limited lifespan, are susceptible to aging in older in vitro cultures, and experience a loss of potential differentiation as the time is increased in culture. One of the most commonly used supplements in mesenchymal stem cell culture is ascorbic acid. This paper describes the role of ascorbic acid against the proliferation and differentiation of mesenchymal stem cells into osteoblasts and chondrocytes in vitro. This paper is a review of literature obtained through Google and Pubmed search. The results obtained that mesenchymal stem cell culture medium supplemented with ascorbic acid will increase the proliferation and differentiation of mesenchymal stem cells into osteoblasts and chondrocytes. Therefore, ascorbic acid can be used for expansion of mesenchymal stem cells in regenerative disease therapy.

Sel punca merupakan sel yang memiliki kemampuan memperbanyak diri (self renewal), belum mempunyai bentuk dan fungsi yang spesifik (undifferentiated) namun memiliki kemampuan untuk berdiferensiasi menjadi sel lainnya. Faktor utama yang menentukan keberhasilan proliferasi dan diferensiasi sel punca mesenkim secara in vitro adalah kondisi kultur. Sel punca mesenkim memiliki masa hidup yang terbatas, mudah mengalami penuaan pada kultur in vitro yang lama, dan mengalami kehilangan potensi diferensiasi dengan meningkatnya waktu dikultur. Salah satu suplemen yang sering digunakan dalam kultur sel punca mesenkim adalah asam askorbat. Tulisan ini menguraikan tentang  peran asam askorbat terhadap proliferasi dan diferensiasi sel punca mesenkim menjadi osteoblas dan kondrosit secara in vitro. Tulisan ini berupa review literatur yang didapatkan melalui pencarian Google dan Pubmed. Hasil yang didapatkan bahwa medium kultur  sel punca mesenkim yang disuplementasi dengan asam askorbat akan meningkatkan proliferasi dan diferensiasi sel punca mesenkim menjadi osteoblas dan kondrosit.  Oleh sebab itu, asam askorbat dapat digunakan untuk ekspansi sel punca mesenkim dalam terapi penyakit regeneratif.
