Abstract

Abstract

Shigella dysenteriae is the leading cause of bacillary dysentery or shigellosis, causing 1.1 million deaths per year. Adhesion is a very important initial stage in the process of infectious disease. This study was aimed to analyze the ability of the Outer Membrane Protein (OMP) Shigella dysenteriae 237 kDA as an adhesive protein for enterocytes of mice BALB/c strain. The research stages included identification of S. dysenteriae isolation of OMP S. dysenteriae, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), hemagglutination test, isolation of enterocyte of BALB/c mice, and adhesion test. Outer Membrane Protein S. dysenteriae 237 kDa is a protein adhesin on enterocytes pf BALB/c mice because it can inhibit the adhesion of S. dysenteriae bacteria to enterocytes of BALB/c mice. The adhesion index was influenced by the OMP hemagglutinin protein concentration at 78.7% and the remaining 21.3% was influenced by other factors. The conclusion from the results of this study was that OMP S. dysenteriae 237 kDa is protein adhesin in enterocytes of BALB/c mice.

Abstrak
Shigella dysenteriae adalah penyebab utama disentri basiler atau shigelosis, penyebab kematian 1,1 juta jiwa per tahun. Adhesi adalah tahap awal yang sangat penting dalam proses terjadinya penyakit infeksi. Penelitian ini bertujuan untuk menganalisis kemampuan protein Outer Membrane Protein (OMP) S. dysentrieae 237 kDa sebagai protein adhesif enterosit mencit galur BALB/c. Tahapan penelitian terdiri dari identifikasi S. dysenteriae, isolasi OMP S. dysenteriae, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), uji hemaglutinasi, isolasi enterosit mencit galur BALB/c, dan uji adhesi. OMP S. dysenteriae 237 kDa merupakan protein adhesin pada enterosit mencit galur BALB/c karena dapat menghambat adhesi bakteri S. dysenteriae pada enterosit mencit galur BALB/c. Indeks adhesi dipengaruhi oleh konsentrasi protein hemaglutinin OMP sebesar 78,7% dan 21,3% sisanya dipengaruhi oleh faktor lain. Kesimpulan penelitian ini adalah OMP S. dysenteriae 237 kDa merupakan protein adhesin pada enterosit mencit galur BALB/c.
