EFEK ANTIHIPERURISEMIA FRAKSI-FRAKSI KAYA FLAVONOID BATANG BROTOWALI (Tinospora crispa) PADA MENCIT MODEL HIPERURISEMIA
Abstract
Brotowali (Tinospora crispa) secara tradisional dimanfaatkan untuk mengobati penyakit gout, artritis reumatoid, dan peradangan internal. Studi bioaktivitas dari batang brotowali menunjukkan efek antioksidan, antinosiseptif, dan antiinflamasi. Penelitian ini bertujuan untuk mengevaluasi efek antihiperurisemia dua fraksi dari ekstrak etanol batang brotowali pada mencit model hiperurisemia akut yang diinduksi potasium oksonat. Batang brotowali diekstraksi dengan etanol 70%, kemudian dipartisi secara berurutan dengan n-heksana, kloroform, dan etil asetat. Analisis fitokimia dilakukan dengan metode kromatografi lapis tipis, sedangkan kadar asam urat serum diukur dengan metode enzimatik-kolorimetri. Studi kami sebelumnya telah melaporkan bahwa ekstrak etanolik dan fraksi tidak larut n-heksana (FHITS) batang brotowali memiliki kandungan flavonoid yang tinggi. Pada studi ini, flavonoid tidak terdeteksi dalam fraksi kloroform (FKTS), namun ditemukan lebih dominan dalam fraksi etil asetat (FETS). Pada penelitian sebelumnya, ekstrak hidroalkoholik 500 mg/kg dan FHITS 100 mg/kg menunjukkan efek antihiperurisemia sebanding dengan allopurinol 10 mg/kg. Perlakuan fraksi kloroform dan fraksi etil asetat masing-masing dengan dosis 100 mg/kg pada mencit model hiperurisemia menunjukkan persentase penurunan kadar asam urat berturut-turut sebesar 39% dan 52%. Dengan demikian, ekstrak batang brotowali atau fraksinya yang mengandung flavonoid berpotensi digunakan dalam pengobatan hiperurisemia.
